Gram Stain Report Requirements
The following is an outline for the report. It will be graded out of a total of 100 points. The report should have appropriate font style and size (10-12), equal to or less than 1-inch margins, appropriate indenting and at least three outside sources. Figures should have a figure number, title, reference. See below for specific information about each individual part of the report and the rubric which will be used for grading.

➢ What are the two major types of bacterial cell walls, what are their major and minor parts and their functions? Should include a figure of both types of cell walls. (20 of 50 points)
➢ Describes what happens to the two types of cells (chemically speaking) at each step of the Gram stain. Should include a figure for better description. (20 of 50 points)
o What happens to each cell type when crystal violet is applied?
o What happens to each cell type as iodine is applied?
o What happens to each cell type as decolorizer/alcohol is applied?
o What happens to each cell type as safranin is applied?
➢ What role does the Gram stain play in microbiology and medicine? (10 of 50 points)
Results (15 points)
➢ This section should include 3 figures:
o a) Gram stain of the Gram positive bacterial culture (S. aureus)
o b) Gram stain of the Gram negative bacterial culture (E. coli)
o c) Gram stain of the mixed bacterial culture (S. aureus and E. coli)
➢ Figures can be hand drawn or from the web (appropriate references cited). Figures should have a figure number, title, reference and should be labelled with arrows appropriately and drawn in the right colors. Include the microscopic observations (color, shape and grouping).
➢ Summarize your results under every figure in a few sentences.
Conclusions (15 points)
➢ From your results, how can you categorize the microbes you view under the microscope?
Which set of observations/characteristics belonged with each organism used?
o What color do each of your bacterial samples appear as?
o Are they Gram positive or Gram negative?
o What are their shapes/arrangements?
➢ How did you come to those conclusions? (tell me what lead you to that thought, connect it back to the principle of Gram Staining)
o Why did each of your bacterial samples stain the particular color and why?
References: (5 points)
➢ Correct reference format must be used (either APA or MLA).
➢ References should be numbered, and the number added to the report when necessary. Spelling of the authors of the references must be correct.

➢ Overall format and quality of lab report, spelling and grammar
➢ Use of proper scientific name formatting in the entire report including references.
➢ Pages numbered, figures numbered, labelled and appropriately referenced in the various
