JB Chasers is a small sports Bar chain in the DFW area. They are known for their Thursday night $5.00 pitchers of beer and the all you can eat calamari bar. You and your group decide to go to JB Chasers for a well-deserved break from Cell Biology class. Your group is seated as soon as you arrive. One of your group has called ahead and ordered beer and the first two plates of calamari so it is ready as soon as you are seated. Everyone is commenting on how good the calamari are, and end up ordering another two plates within 15 minutes. As everyone starts to devour the next two plates of calamari one of your group says they were reading an article and it says that someone was suing JB Chasers for substituting pork intestines for calamari. Every one spits the calamari into a napkin! Apparently pig intestines and calamari have the same texture when fried and pig intestines are much cheaper than squid. One of your group realizes that the techniques you learned in Cell Biology would be helpful in determining wither or not you have been purchasing real squid or Pig intestines. Would be nice to know if you were getting ripped off or no. Your group takes home some of the calamari and freeze’s it so you can take it to the lab on Monday.
To answer this question, you need to review all of your knowledge from this semester, the paper you just turned in, and those circular appendages on top of your neck.
You are being asked to wright at least two pages describing how and why you would approach this issue. For instance, I have “calamari” my known from JB Chasers. I would need to get some pork intestine and squid from the store for comparison. Would also need maybe a fish, octopus and beef to help place the samples on a cladogram if one is needed. Do you need those or just the pork and squid?
Think like a scientist and lay out a plan using all the techniques you learned this semester. Tell me how you will proceed. Be as detailed as possible in the two pages you have.
Framework for The Answer: Your answer should address five key areas (a) samples utilized and why, (b) sample preparation from raw sample to isolated protein (ready to load), including the major steps involved and the purpose of each, i.e. Using the DeNovix and why (c) protein separation – describe how you are going to separate the proteins and why (what instruments are being used, what steps allowed you to visualize the proteins, (d) data analysis – now the proteins are separated, what are you looking to find and how will you find it; and (e) possible results and conclusion – describe and explain what you would observe if the sample from JB Chasers is squid/calamari? If it was not? Why is this important and what would you do with this information?
