according my mails instruction is,
Everything you need to begin is in your lab manual. The list that your unknowns come from is on page 15 of the lab manual ” List of Possible Bacteria.The actual description of the project can be found on pages 117-126. You will need to read over this very carefully and look up A LOT in your lab manual especially the significance of selective/differential test results and biochemical test results.read the pages on 117 -126 over and look at the streak plate results and the gram stain results for both of your unknowns. From this information try to narrow down the list on page 15,It is to just help pace you with this project.
the Selective and Differential Media Test Results for the unknown bacteria. They are located right under the Gram Stain Results. You could have been able to narrow down your unknown from the list in your lab manual on page 15. Now, the results you will see for the differential and selective media plates have been chosen for you because these give some of the most significant results specific for your unknowns. Each Unknown has 5 media. Keep in mind the NA is Nutrient Agar and is your control. You will have to use your lab manual to figure out the significance of the results. This will take some time. Don’t get frustrated. Keep is well organized.
I will not be collecting lab notebooks for this project for obvious reasons, so everyone will get these points automatically.
Please read it carefully. There are two things that you are not responsible for in this project. First is the Lab Notebook, and second is Making the Dichotomous Key that starts on page 119 and ends on page 120. You are NOT responsible for either of those things.
this Unknown Lab project, will be heavily dependent upon the tables and charts in your lab manual, specifically those for the selective/differential media tests and the biochem tests.
Remember, one of your unknowns is gram positive and the other is gram negative (the lab manual tells you this as well). Depending on what your last name starts with you either have #1, and #3 OR #2 and #4. (Every student has 2 unknowns).
***HINT: #1 and #2 are Gram (-), and #3 and #4 are Gram (+). Remember. They are pictures.

NOW, go to page 18 with the information for each unknown and narrow the list down. You should make a column for each unknown and write the possible bacteria for each under them. Keep your info well organized.

List the tests and write down the result and the Presumptive ID for each using the tables on page 87-95. You will need to read the background information carefully and look up anything you may not understand in your text book as well. Pay special attention to MSA, PEA and Blood. You should know what each of these tests tell you about your unknown.

List the tests and write down the result and the Presumptive ID for each using the tables on page 87-95. You will need to read the background information carefully and look up anything you may not understand in your text book as well. Pay special attention to HEK and Endo. You should know what each of these tests tell you about your unknown.
Although there are more tests you did these are key ones to focus on that will certainly help you narrow your list further.
*NOTE: NA, nutrient agar is the control. You should know what it means to have a control in an experiment like this!
Most important to me than the correct identification of the unknowns is your logic in determining which unknowns you came up with. You need to support your answer with evidence from your results. Another way to put this is that you need to strongly support the answer to the question WHY? Do you think #1 is ____________ for example.
The last piece of information I am providing attached below is from Bergey’s Manual. This is the “encyclopedia” of bacteriology. I have scanned the pages that you would need for these unknowns.
DO NOT dwell on results that do not correspond to what you think it is. For example if one of the test results like VP is different from what you have for your unknown compared to what is in Bergey’s for the bacteria you think you have, chalk it up to error. You are looking for the majority of your tests to correspond or “match”.
You should ONLY use this manual at the very end of your findings. Otherwise it may confuse you further if you have not followed the steps I suggested. Look at the Project outline on page 119. It is listed in 5 weeks, but I gave you all of these results in 3 weeks. AND AGAIN, you are not responsible for doing the Dichotomous Key.
